Scientific Overview & Physiological Context

Marek's Disease is a lymphoproliferative herpesvirus infection of chickens that causes cancerous T-cell tumors (lymphomas) in internal organs, asymmetric nerve paralysis, and lifelong viral shedding in feather dander.

How the Virus Causes Cancer: The Meq Oncogene

Marek's Disease Virus (MDV-1) is an Alphaherpesvirus with a unique weapon: a cancer-causing gene called Meq. After a chick inhales the virus from infected dust, the virus replicates in B cells and then invades CD4+ T lymphocytes. The Meq gene turns these healthy T cells into malignant cancerous lymphoma cells that aggressively infiltrate the liver, spleen, kidneys, heart, and skin.

Sciatic Nerve Swelling & The Split-Leg Posture

One of the most famous clinical signs of Marek's disease is progressive leg or wing paralysis. Infiltrating lymphoma cells swarm into the major nerve trunks, especially the sciatic and brachial nerves. The affected nerve swells to two or three times its normal thickness and loses its clean, cross-striated appearance. The bird is unable to walk, adopting the classic split-leg posture with one leg stretched forward and the other backward.

The 'Leaky Vaccine' & Liquid Nitrogen Handling

Marek's disease vaccines (like CVI988/Rispens and HVT) are 'leaky'—they protect the bird against developing cancerous tumors and paralysis, but they do not stop the bird from catching the virus or shedding it into the litter. Because CVI988 vaccine is made of living host cells, it must be stored in liquid nitrogen at -196°C. Thawing ampoules too quickly or letting diluent warm above 25°C will destroy the live cells and cause complete vaccine failure.

Veterinary Takeaway & Practical Application

Marek's disease vaccine must be handled with extreme care. Keep vaccine ampoules submerged in liquid nitrogen (-196°C) until the moment of dilution, and administer the reconstituted vaccine within 2 hours to ensure maximum cell viability.
